De film begint vrij heftig: Naveen wordt tegen hun wil in vastgehouden in India als hun ouders achter hun gender en seksuele oriëntatie komen. Later komen persoonlijke groei, bevrijding en oprecht geïnteresseerde en liefhebbende schoonouders voorbij.

MAJOR! vertelt over het leven en de strijd van Miss Major Griffin-Gracy, een 73-jarige zwarte transgender activist die al veertig jaar vecht voor de rechten van gekleurde transvrouwen. Miss Major is een veteraan van de Stonewall opstand, een overlevende van de Attica staatsgevangenis, een voormalige sekswerker, een community leader en mensenrechtenactivist.
Haar persoonlijke verhaal is verstrengeld met de LHBTQ-strijd voor de rechtvaardigheid en gelijkheid van de jaren ’60 tot vandaag.
